Enhanced Resistance of Bone Marrow Transplanted Mice to Bacterial Infection
نویسنده
چکیده
The in vivo effect of recombinant murine granulocytemacrophage colony stimulating factor (rGM-CSF) on the resistance of mice to bacterial infection and on the number and function of neutrophils was studied in lethally irradiated mice transplanted with syngeneic bone marrow cells. Bone marrow transplanted (BMT) mice were injected intraperitoneally with 150 ng rGM-CSF or buffer solution (diluent) twice daily for 18 consecutive days. Total neutrophil recovery from the peripheral blood and the number of neutrophils mobilized into the peritoneal cavity were accelerated in rGM-CSF-treated recipients. Peritoneal neutrophils isolated from mice treated with rGM-CSF exhibited primed superoxide generation (02-) after in vitro stimulation with suboptimal concentrations of phorbol myristate acetate (PMA), as compared with control mice (treated with diluent). No additional increase in 0,production

متن کاملHost resistance to Listeria monocytogenes infection is enhanced but resistance to Staphylococcus aureus infection is reduced in acute graft-versus-host disease in mice.
Acute graft-versus-host disease (GVHD) is characterized by the production of high levels of T helper 1 (Th1)-type cytokines. Bone marrow transplantation from allogeneic C57BL/6 cells to CBF(1) mice produced acute GVHD. Host resistance to Th1-driven Listeria monocytogenes was enhanced, whereas host resistance to Th2-driven Staphylococcus aureus was reduced during acute GVHD. These results sugges...
متن کاملTherapeutic Potential of Mouse Bone Marrow Mesenchymal Stem Cells in Carbon Tetrachloride (Ccl4)-Induced Liver Fibrosis
Purpose: To study the effect of allogenic bone marrow mesenchymal stem cells (BMMSCs) transplantation on carbon tetrachloride-induced liver fibrosis in mice. Materials and Methods: Fifty five female NMRI mice were divided in 5 groups, and to induce liver fibrosis CCL4 intraperitonealy was injected 1ml/Kg twice a week for 8 weeks 106 allogenic BMMSCs were infused in cell therapy group via tail v...
متن کاملAutophagy-Modulated Human Bone Marrow-Derived Mesenchymal Stem Cells Accelerate Liver Restoration in Mouse Models of Acute Liver Failure
Background: Mesenchymal stem cells (MSCs) have been recently received increasing attention for cell-based therapy, especially in regenerative medicine. However, the low survival rate of these cells restricts their therapeutic applications. It is hypothesized that autophagy might play an important role in cellular homeostasis and survival. This study aims to investigate the regenerative potentia...
متن کاملMembrane TNF confers protection to acute mycobacterial infection
BACKGROUND Tumour necrosis factor (TNF) is crucial for the control of mycobacterial infection as TNF deficient (KO) die rapidly of uncontrolled infection with necrotic pneumonia. Here we investigated the role of membrane TNF for host resistance in knock-in mice with a non-cleavable and regulated allele (mem-TNF). METHODS C57BL/6, TNF KO and mem-TNF mice were infected with M. tuberculosis H37R...
